Circulation Research is available at http://circres.ahajournals.org DOI: 10.1161/CIRCRESAHA.116.308391 Abstract: Growth differentiation factor 11 (GDF11) and myostatin (or GDF8) are closely related members of the transforming growth factor β superfamily and are often perceived to serve similar or overlapping roles. Yet, despite commonalities in protein sequence, receptor utilization and signaling, accumulating evidence suggests that these 2 ligands can have distinct functions in many situations. GDF11 is essential for mammalian development and has been suggested to regulate aging of multiple tissues, whereas myostatin is a well-described negative regulator of postnatal skeletal and cardiac muscle mass and modulates metabolic processes. In this review, we discuss the biochemical regulation of GDF11 and myostatin and their functions in the heart, skeletal muscle, and brain. We also highlight recent clinical findings with respect to a potential role for GDF11 and/or myostatin in humans with heart disease. Finally, we address key outstanding questions related to GDF11 and myostatin dynamics and signaling during development, growth, and aging. (Circ Res. 2016;118:1125-1142. Growing backwards: an inverted role for the shrimp ortholog of vertebrate myostatin and GDF11.
Myostatin (MSTN) and growth differentiation factor-11 (GDF11) are closely related proteins involved in muscle cell growth and differentiation as well as neurogenesis of vertebrates. Both MSTN and GDF11 negatively regulate their functions. Invertebrates possess a single ortholog of the MSTN/GDF11 family. In order to understand the role of MSTN/GDF11 in crustaceans, the gene ortholog was identifi...
متن کاملDifferential antagonism of activin, myostatin and growth and differentiation factor 11 by wild-type and mutant follistatin.
Follistatin binds and neutralizes members of the TGFbeta superfamily including activin, myostatin, and growth and differentiation factor 11 (GDF11). Crystal structure analysis of the follistatin-activin complex revealed extensive contacts between follistatin domain (FSD)-2 and activin that was critical for the high-affinity interaction. However, it remained unknown whether follistatin residues ...
متن کاملLifelong exercise, but not short‐term high‐intensity interval training, increases GDF11, a marker of successful aging: a preliminary investigation
Lifelong exercise is associated with regulation of skeletal mass and function, reductions in frailty, and successful aging. Yet, the influence of exercise on myostatin and myostatin-interacting factors is relatively under examined in older males. Therefore, we investigated whether serum total myostatin, free myostatin, follistatin, and growth and differentiation factor 11 (GDF11) were altered f...
متن کاملThe Immateriality of Circulating GDF11.
Recent portrayals of growth differentiation factor (GDF)-11 as an endocrine fountain of youth fail to consider the competitive influences of myostatin, a homologous protein with true endocrine action that binds the identical receptor, activin receptor IIb (ActRIIb). Subsequent studies have disproven the original premise that circulating levels of GDF11, but not those of myostatin, decline with ...
متن کاملQuestions and Answers About Myostatin, GDF11, and the Aging Heart.
M yostatin and GDF11 are highly homologous transforming growth factor (TGF)-β family members implicated in skeletal and cardiac muscle growth. Myostatin promotes muscle wasting. In contrast, GDF11 was identified as a factor that reverses aging-related cardiac hypertrophy, and in separate experiments, GDF11 supplementation was linked to the reversal of aging-associated skeletal muscle dysfunctio...
